RADIATION BIOLOGY


CPRM has an active Radiation Biology Research Program. A major area of focus is on tumor radio-resistance and radio-sensitization. In terms of radio-resistance, an active area of research is on cell signaling pathways involving the K-RAF signaling axis. Our group has demonstrated that non-canonical, kinase independent functions of RAF mediates cell cycle progression and radio-resistance through phosphorylation of CRAF at serine 338 by PAK1. These results provide multiple potential avenues for clinical trials to enhance tumor control.

Our group is also developing tumor targeted delivery approaches of ultra-potent tumoricidal drugs that have the dual advantage of also radio-sensitizing. We were the first group to identify that the anti-tubulin drug monomethyl auristatin E (MMAE) is a potent radio-sensitizer when compared to clinically used radio-sensitizers such as cisplatin or taxanes.


Our Radiation Biology Group is also focused on integrating Radiobiology and Bioengineering innovations to develop small molecules, specialized nanoparticles, and devices, to enhance tumor control, reduce normal tissue toxicity, and promote recovery of radiotherapy-damaged tissues.    Specific disease areas include tumors of the brain, head and neck, and pancreas, and radiotherapy induced brain and heart injury.

In this general context our current translational and mechanistic investigations are directed towards, (1) a radiation activated nanoparticle based prodrug system coupled with radio- and brachytherapy machines, (2) a tumor selective small molecule radio/chemosensitizer, and (3) a novel device and targeted nanoparticles to stimulate and support localized tissue recovery.
